Viability and taxonomic confirmation of cultures of Trichophyton tonsurans (Malmsten) preserved under mineral oil

The fungi are heterotrophic, eukaryotes, and cosmopolitan, since they are present in any part of the planet. Being widely distributed in nature, are found in water, air, soil, plants and humans and can cause disease. Dermatophytes are fungi that colonize the keratinized tissue of the skin, hair and fingernails of humans and other animals. Trichophyton tonsurans is an anthropophilic dermatophyte and can cause especially tinea capitis. Numerous techniques are used to maintain and most suitable fungus is one that lasts for long periods of culture unique features, such as viability, sporulation and pathogenicity. In order to investigate the viability and confirm the taxonomy of the fungus Trichophyton tonsurans colonies, 69 isolates were analyzed, all the URM Culture Collection - UFPE stored under mineral oil. Fragments of the cultures were reactivated dextrose broth, kept at 28 ° C ± 2 ° C for 12 days. Of the 69 samples analyzed, 42 (60.87%) are viable after the storage period 1955-2008. The colonies that showed growth in glucose broth were transferred to Sabouraud ágar, staying for 08 days at room temperature (RT). After growth, the colonies were on Sabouraud agar culture technique referred to slide to facilitate confirmation process Taxonomy. Forty-two samples had confirmed the taxonomy, with a total percentage of 100%. Thus, it was found that the preservation method in mineral oil is recommended for T. tonsurans after long storage periods.
