Pathogenic characterization of Pestalotiopsis grandis-urophylla by mycelium agar plugs inoculation

Abstract

The aim of this study was to carry out the pathogenic characterization of Pestalotiopsis grandis-urophylla isolates obtained from lesions on eucalyptus leaves. From 2 isolates, pathogenic characterization was carried out, where they were inoculated in leaves and subjected to controlled conditions of temperature and light in transparent acrylic boxes with maintenance of humidity only on paper. The evaluations were carried out by measuring the lesions on the abaxial surface of the leaves at 4, 6, 8 and 10 days after inoculation (DAI), with the aid of a digital caliper, obtaining the size of the lesions in mm2. In assessing the pathogenic potential, data referring to lesions caused by P. grandis-urophylla were submitted to Tukey's test and regression analysis to obtain significant models. There was no statistical difference between the isolates regarding the injured leaf area (ILA) and the area under the disease progress curve (AUDPC). The evaluated inoculation methodology provided reproduction of disease symptoms.
